The Mt Glorious course climbs from the Cedar Flats valley up a steep ridgeline firebreak, then drops to England Creek before a long, continuous climb to the summit of Mount Glorious. The final stretch follows a historic stone-lined western ridge single trail through rainforest, with stone stairs leading to the Western Window and a flat run to the finish. Much of the route is shaded by trees, but the racing day in November can turn hot, so runners should expect to carry water and sun protection. In 2026 the race also hosts the Queensland Mountain Running Championships, held in conjunction with Queensland Athletics, and a shorter 11km race is offered alongside this 22km event.

Much of the course is on the west side of the ridge, where trees provide shade for much of the route. Even so, the day can be hot, so suitable clothing and sunscreen are recommended.
